1240 Distinct cAMP signaling microdomains differentially regulate melanosomal pH and pigmentation

چکیده

cAMP signaling is well-established in the regulation of melanin synthesis. Two distinct pathways, melanocortin 1 receptor (MC1R) and soluble adenylyl cyclase (sAC) are known to differentially modulate melanogenesis. The MC1R pathway control melanogenesis via gene expression, while sAC influences synthesis melanosomal pH. However, whether affects pH remained unclear. Using human mouse melanocytes lacking signaling, we show that loss does not alter Further, require active either or melanocytes. Thus, appears be only involved regulation. Since function stimulates wild type melanocytes, also asked if sAC-dependent found has no effect on skin hair content null (e/e) mice. Interestingly, application forskolin, which activates downstream increase epidermal eumelanin e/e mice, leads greater enhancement production knockout mice relative MC1R- pathways define mechanisms regulate pigmentation.

Different phosphodiesterases (PDEs) regulate distinct phosphoproteomes during cAMP signaling.

Sixty years ago, Sutherland and Rall identified cAMP as the first second messenger involved in cell–cell communication (reviewed in ref. 1). For many years, Sutherland had been interested in understanding how hormones, such as catecholamines, released by one cell type could alter the characteristics of a target cell because hormones of this type were charged, and therefore could not enter the c...

Distinct cAMP signaling pathways differentially regulate alpha2C-adrenoceptor expression: role in serum induction in human arteriolar smooth muscle cells.

The physiological role of alpha(2)-adrenoceptors (alpha(2)-ARs) in cutaneous, arteriolar, vascular smooth muscle cells (VSMs) is to mediate cold-induced constriction. In VSMs cultured from human cutaneous arterioles, there is a selective increase in alpha(2C)-AR expression after serum stimulation. In the present study, we examined the cellular mechanisms contributing to this response. Serum ind...

cAMP signaling microdomains and their observation by optical methods

The second messenger cyclic AMP (cAMP) is a major intracellular mediator of many hormones and neurotransmitters and regulates a myriad of cell functions, including synaptic plasticity in neurons. Whereas cAMP can freely diffuse in the cytosol, a growing body of evidence suggests the formation of cAMP gradients and microdomains near the sites of cAMP production, where cAMP signals remain apparen...

Bone morphogenetic proteins differentially regulate pigmentation in human skin cells.

Bone morphogenetic proteins (BMPs) are a large family of multi-functional secreted signalling molecules. Previously BMP2/4 were shown to inhibit skin pigmentation by downregulating tyrosinase expression and activity in epidermal melanocytes. However, a possible role for other BMP family members and their antagonists in melanogenesis has not yet been explored. In this study we show that BMP4 and...

Role of Membrane Microdomains in Compartmentation of cAMP Signaling

Spatially restricting cAMP production to discrete subcellular locations permits selective regulation of specific functional responses. But exactly where and how cAMP signaling is confined is not fully understood. Different receptors and adenylyl cyclase isoforms responsible for cAMP production are not uniformly distributed between lipid raft and non-lipid raft domains of the plasma membrane. We...
